In vitro REGENERATION USING BULB AND LEAF EXPLANTS OF Urginea indica Kunth

Abstracts

Urginea indica, a medicinally important plant need to be conserved for its ethno botanical importance and for future innovations in medicine. In vitro techniques were employed to multiply and conserve this medicinally valued plant. Bulb and leaf explants were cultured on Murashige and Skoog’s medium supplemented with various growth regulators. Callusing, direct and indirect regeneration from the cultures were observed depending on the media composition. MS+2,4D (13.56 µM) and MS+NAA (16.11µM) were found to be best for callusing, while MS+KIN (18.6µM) was found a conducive medium for direct regeneration of bulb explants. Maximum regeneration potential of shoots was exhibited by bulb explants on MS+KIN (13.95µM) +2, 4-D (6.78µM) +GA3 (2.88µM) medium through indirect organogenesis when 40 days callus cultured on auxin medium. The medium constituting ½ MS+IBA (2.45 µM) was suitable for rooting. In vitro raised plants were better hardened in perlite mixture. Fully acclimatized plants were maintained in poly house and about 87% survival was recorded.
